I almost always start with locate -i
Only after I get feedback from that--I'll sometimes use `locate` without the case insensitive flag. Is there a config file? Probably not. I didn't find one. locate -i usually tells me what I need to know.

Use an alias, like ilocate (or shorter iloc) for the case insensitive one.
There is no config file for options in either slocate or mlocate, I do not know which one you're using (or even the old GNU locate?).

The locate command is not configurable in the sense that it is what it is, but there are many arguments you can use to tailor it to particular situations. See man locate for the list.
If there's a particular set of arguments you need to use regularly, one option may be to write a simple script ("mylocate.sh"?) including those arguments and call the script when you need to use that particular version of the command.
